3 × 500 µl 10× Ligation BufferThe biotechrabbit T4 DNA Ligase, Rapid, is an exceptionally pure highly concentrated ligase for applications where high concentrations of the enzyme are required. It is especially recommended for fast ligations.
T4 DNA Ligase, Rapid is supplied with two buffers – the common 10x ligation buffer for usual 1 hour or overnight ligations and the 2x Rapid Ligation Buffer containing PEG for fast 5- 10 minutes ligations or ligation of low concentration or blunt ended DNA.
The 10X Ligation Buffer does not contain PEG and is compatible with standard ligation protocols which do not specify the use of a rapid/fast/quick format buffer.
The T4 DNA Ligase, Rapid in combination with the 2X Rapid Ligation buffer greatly stimulates the rate and efficiency blunt-end ligation, therefore long incubations (>10 minutes) are not recommended as they reduce the transformation efficiency of ligation products.
T4 DNA Ligase catalyzes the formation of a phosphodiester bond between the terminal 5′ phosphate and a 3′ hydroxyl groups of duplex DNA or RNA. The enzyme efficiently joins blunt and cohesive ends and repairs single stranded nicks in duplex DNA, RNA or DNA/RNA hybrids (1).
The enzyme is purified from a recombinant E. coli strain carrying the cloned T4 DNA Ligase gene.
